Cilic hit 11 aces and only dropped two first-serve points in notching his 35th win of 2014.
Cilic, seeded third, owns a 3-0 record against Rosol, including a victory in Rotterdam earlier this season, but the pair have never met on clay.
In other action in Umag, second-seeded Tommy Robredo defeated fellow Spaniard Albert Ramos-Vinolas, 6-4, 6-4.
Robredo, 32, is the defending champion at Umag, while Cilic is the 2012 champion.
On Thursday the top half of the draw, led by last year’s runner-up, top-seeded Fabio Fognini, will make their bids for the quarterfinals. Local fans will be closely watching 17-year-old Croatian Borna Coric. Coric won his first ATP main draw match on Monday in Umag, and will face Horacio Zeballos. Sixth-seeded Andreas Seppi and fourth-seeded Joao Sousa will also be in action.
